Site-selective protein functionalization serves as an invaluable tool for investigating structures and functions in complicated cellular environments accomplishing semi-synthetic conjugates such traceable therapeutics with improved features. Dual of proteins allows the incorporation two different types functionalities at distinct location(s), which greatly expands features native proteins. The attachment crosstalk a fluorescence donor acceptor dye provides fundamental insights into folding structural changes upon ligand binding their environments. Moreover, combination drug molecules modes action, imaging agents or stabilizing polymers new avenues to design precision reproducible well-characterizable fashion. We report the facile preparation of palladacycles as storable arylpalladium(II) reagents from acetanilides via cyclopalladation. The palladacycles exhibit good stability in PBS buffer and are capable of functionalizing a metabolically encoded HPG-containing protein, thus providing a new type of biocompatible organometallic reagent for selectively functionalizing the alkyne-encoded proteins.
